Go to the Dimension tool and hover over first hole to highlight its axis. Once highlighted, select the axis and then hold down CTRL to select the axis located at center of the part on which the holes are created. While still holding CTRL, hover over the second hole and select its axis. The dimension should now show the angle between holes.  You can later hide the points in the structure tree if you want to.

Does this work on existing geometry?

No you will have to recreate and record the steps for the operations to parameterize.
