Is this how your window looks after you load the .aedt model?
Is there any error message in the message manager window?

Here is some information.
1).Need administrator rights
2). Set the Ansys Path in MATLAB. For example, In Home-> Set Path -> Add ÔÇ£C:\Program Files\AnsysEM\AnsysEM21.2\Win64\cpl\matlab\r2020aÔÇØ, that is AnsysEM installation folder.
Now, Twin builder/Simplorer 2021R2 does not support 2020b or later.
Modifying startup.m is one variation of the option of setting path, and it is not necessary after already adding path. To avoid misleading something, suggest checking if path is correct. Please delete it in startup.m file if the path is not correct.
There is more information on it in the manual of 2021R2,. Please review it in Twin Builder Help > Product Coupling > Simulink® SubCircuits > Program Requirements for MATLAB®/Simulink® SubCircuits.
3). Follow steps in the manual, to add Simulink components in Simplorer model.
i). Have Matlab/Simulink model and add Simulink component.
ii). Add pins in the Simulink component of Simplorer model. Assign those pins with Simplorer modelÔÇÖs SDB name. Adding pins is required.
Simulink modelÔÇÖs name should be shown in Project manager tree of AEDT model. We could double click to open the Simulink model linked to Simplorer.
4). Add S-function in linked Simulink file. The Simplorer model should be loaded in the setting dialog automatically. ÔÇ£LinkÔÇØ port names in the dialog of "link assignment".
5). Run co-simulation from either Simulink or Simplorer.
