Ansys EmployeeInterference command identifies interfering portion between two bodies and substracts it from either smaller or bigger body based on option selected. So at the end of Interference command, there will be two resulting bodies, one with original size and second with interference volume subtracted size.
Whereas, Overlap Bodies command also identifies overlapping portion between two bodies and does boolean operations. But at the end of Overlap Bodies command , there will be three resulting bodies, two original bodies with overlapping volume subtracted from each of them and a body with overlapping volume. -

Ansys EmployeeYes, if there are interference in the assembly, you should use Interference tool and remove them before share topology -

Ansys EmployeeBy default Spaceclaim substracts from the larger body. But you select option 'Substract from smaller body'
