is it somehow possible to activate Discovery license on shop floor machine without internet connection?

You need to be connected to the internet while activating the Discovery subscription license for the first time. Once the subcription gets activated, you can use it without an internet connection for 30 days. You need to connect to the internet and refresh the license connection every 30 days.

Is there a way to register a pc offline that cannot connect to the internet because of security reasons?

With subscription license, offline activation is not possible. You may want to check out the floating license option which can be installed on a server machine and doesn't need internet for activation.
