Please see the link below on how to create a mixture. (section 16.1.4 of Ansys Fluent 2020 R1 User Guide)
Once the mixture is created, at the inlet boundary, you can define the mass fraction of each of the species.Â

SubscriberSir, if in a simulation Im filling a room with a gas through an inlet, does the room interior contains air as default before letting gas inside in ansys fluent?? or i should assign interior fluid as air?

You need to assign the material for the cell zone. In the 'Physics' tab, under 'Zones', select 'Cell Zones' and ensure that the Material Name is set to whatever material you want inside this cell zone.Â
